چکیده

The name Cholamonas cyrtodiopsidis gen. n., sp. n., is created for a mycophagous heterotrophic flagellate isolated and cultivated from the intestine of a diopsid fly. Flagellates were subanteriorly biflagellate, uninucleate, and naked. Golgi dictyosomes were anterior to the nucleus, and a reticulate paranuclear body posterior to it. Two groups of refractile bodies were present, one at the anterior end of the cell, the other in the vicinity of the nucleus. Numerous elongate, unbranched mitochondria with tubular cristae were distributed around the periphery and parallel to the long axis of the cell. The kinetid consisted of two symmetrical subunits, each with two basal bodies (one of which was associated with a stubby flagellum), a compound microtubular root, and associated fibers and bands. Cytoskeletal microtubules emanated from the cell anterior and were not associated with any kinetid element. Feeding, on yeast, was accomplished by rapid pseudopodial action at the posterior end of the cell. Cholamonas cyrtodiopsidis is referred to Cercomonadida because it possesses a paranuclear body and has a kinetid architecture similar to some species of Cercomonas. It differs from all other cercomonads in its endocommensal habitat, mycophagy, doubled kinetid, distribution of refractile granules and mitochondria, and minimal production of pseudopodia.

Factors related to heterotrophic bacterial and flagellate abundance in temperate rivers

Thirty-one rivers of varying trophic status located in Ontario and Quebec, Canada, were sampled in order to determine the factors related to heterotrophic bacterial and flagellate abundance. Bacterial abundance ranged from 1.80 X 106 to 9.92 X 106 cells ml-' and flagellate abundance ranged from 1.18 X 103 to 17.4 X 103 cells ml-' There was a highly significant, positive relationship between bac...

Fatty acid composition of the heterotrophic nanoflagellate Paraphysomonas sp. : influence of diet and de novo biosynthesis

We compared the relative importance of dietary factors versus de novo synthesis in determining the polyunsaturated fatty acid (PUFA) composition of the heterotrophic nanoflagellate Paraphysomonas sp. The flagellate was fed with different mutants of the picocyanobacterial strain Synechocystis PCC6803, which differ in their capability to synthesize specific PUFAs. The desA, desB, and desD genes o...

Heterotrophic Nanoflagellate, Paraphysomonas imperforata

Two different psychrophilic types of the heterotrophic nanoflagellate Paraphysomonas imperforata were isolated from Newfoundland coastal waters and the Arctic Ocean. When fed bacteria without food limitation, both isolates were able to grow at temperatures from -1.8 to 20°C, with maximum growth rates of 3.28 day-' at 15°C and 2.28 day-l at 12.3°C for the Newfoundland and the Arctic isolates, re...

A comparison of the growth kinetics of six marine heterotrophic nanoflagellates fed with one bacterial species

The growth kinetics of 6 species of marine heterotrophic nanoflagellates (5 to 13 pm) were compared. The maximum spec~fic growth rates (p,,,) of the flagellates ranged from 0.035 to 0.21 h ' and the half-saturation constants (K,) ranged from 1.1 to 45 X 106 bacterial cells ml-' Theoretical threshold concentrations were calculated for each flagellate and the values of these generally ranged from...

Development and application of a most-probable-number-pcr assay to quantify flagellate populations in soil samples.

This paper reports on the first successful molecular detection and quantification of soil protozoa. Quantification of heterotrophic flagellates and naked amoebae in soil has traditionally relied on dilution culturing techniques, followed by most-probable-number (MPN) calculations. Such methods are biased by differences in the culturability of soil protozoa and are unable to quantify specific ta...
